Reaction Conditions:

Stage #1: 3,6-dichloropyridazin-4-aminewith sodium hydroxide in water; for 21 h;Heating / reflux;
Stage #2: with acetic acid in water at 20; pH=4;

A mixture of 40 (0.127 g, 0.774 mmol; CASRN 823-58-5) and 4% aqueous NaOH solution (3 mL) was heated at reflux for 21 h, then cooled to RT. The pH was adjusted to 4 with glacial HOAc, then the mixture was partitioned between H2O (20 mL) and EtOAc (30 mL). The aqueous layer was extracted with EtOAc (30 mL). The combined organic layers were dried (MgSO4), filtered, and concentrated to give 0.082 g (73%) of 42a as a pink solid that was used without further purification.
